ラパ・ヌイの土から生まれた老化研究の鍵

最近の携帯は、本当に機能が多いですよね。電源が入っている時は常に会話も聞いていて、リスティング広告に参考にされているとのまことしやかな噂もあります。まあ念のためですが、

「iPhoneの設定 → プライバシーとセキュリティ → マイク
で、不要なアプリのマイク許可を切る。次に、設定 → プライバシーとセキュリティ → トラッキングで、アプリからの追跡要求を制限する。」

という作業は推奨されてますので、おすすめします。

とはいえ、昔の写真が時々アップされるのは楽しみでもあります。

僕のFacebookの背景にも使っていますが、南太平洋に浮かぶ孤島、イースター島。現地名でラパ・ヌイに行ったのは、ちょうど18年前だと写真が上がってきました。懐かしいですね。

多くの人にとって、この島の記憶は、巨大なモアイ像と結びついていると思います。ところが、医学と生命科学の世界から見ると、ラパ・ヌイにはもう一つ、驚くべき物語があります。

この島の沼の土壌から、ある微生物が見つかりました。まさにグレーの服を着た僕の背景にある沼です。そして、その微生物から単離された物質が、島の名を取って「ラパマイシン」と名づけられました。

最初は、カビに効く抗真菌薬として発見された物質でした。ところがその後、ラパマイシンには免疫抑制作用があることが分かり、臓器移植後の拒絶反応を抑える薬として使われるようになります。一般名はシロリムス。ここまでは、薬としての物語です。

しかし、ラパマイシンの本当の面白さは、さらにその先にあります。

それが mTORです。

mTORは、細胞にとっての栄養センサーであり、成長スイッチのような存在です。アミノ酸、インスリン、成長因子、エネルギー状態などを読み取り、細胞が「成長するべきか」「修復に回るべきか」「オートファジーを起こすべきか」を決めています。

つまりラパマイシンは、単なる抗真菌薬でも、単なる免疫抑制薬でもなく、「細胞が時間をどう使うか」を考える入口になったのです。
老化研究においても、ラパマイシンは大きな転換点を作りました。動物実験では、高齢期に入ってから投与しても寿命が延びる可能性が示され、「老化が進んだ後でも、生物学的な介入余地があるのではないか」という考え方を広げました。

ただし、ここは誤解してはいけません。

ラパマイシンは、サプリメントではありません。若返り薬として気軽に使うものでもありません。免疫抑制作用を持つ薬剤であり、感染リスク、脂質代謝異常、口内炎、創傷治癒遅延など、注意すべき副作用があります。

けれども、この薬が示した概念は非常に大きい。

老化は、単なる時間の経過ではない。
栄養、成長、修復、炎症、免疫、代謝が絡み合った、生物学的なプロセスである。

そして、その一部はシグナルとして読み解けるかもしれない。

思えば、モアイ像が沈黙のまま海を見つめているその足元に、生命の時間を制御する鍵が眠っていたという事実そのものが、科学の美しさを物語っています。

ラパマイシンは、ラパ・ヌイの土から生まれ、抗真菌薬として発見され、免疫抑制薬として臨床に入り、そしてmTORを通じて老化研究の地図を書き換えました。

まさに、孤島から始まった生命科学の大航海。老化とは、運命ではなく、シグナルである。そしてシグナルであるならば、いつか人類は、それを読み解き、調律する日が来るのかもしれません。

The Key to Aging Research Born from the Soil of Rapa Nui

Modern smartphones really do have an astonishing number of functions these days. There are persistent rumors that, whenever the power is on, they are constantly listening to conversations and using that information to optimize targeted advertising. Just to be safe, I often recommend the following:

“On your iPhone, go to
Settings → Privacy & Security → Microphone,
and disable microphone access for unnecessary apps.

Then go to
Settings → Privacy & Security → Tracking,
and limit app tracking requests.”

That said, I do enjoy how old photos occasionally resurface.

As some of you may know from the background image on my Facebook page, I once traveled to the remote island floating in the South Pacific known as Easter Island — or, by its local name, Rapa Nui. Recently, my phone reminded me that the trip was exactly eighteen years ago. It brought back wonderful memories.

For most people, memories of this island are inseparably tied to the giant Moai statues. Yet from the perspective of medicine and life sciences, Rapa Nui carries another extraordinary story.

From the soil of a marsh on this island — in fact, the very marsh visible behind me in the photograph where I am wearing gray — researchers discovered a microorganism. From that microorganism, a compound was isolated and named “Rapamycin,” after the island itself.

At first, rapamycin was identified as an antifungal agent.

Later, however, scientists discovered that it also possessed immunosuppressive properties, leading to its use in preventing organ transplant rejection. Clinically, it became known as Sirolimus.

That alone would already make for a remarkable pharmaceutical story.

But the truly fascinating part of rapamycin lies beyond that.

It leads us to mTOR.

mTOR functions as a nutrient sensor and growth regulator within cells. It interprets signals such as amino acids, insulin, growth factors, and cellular energy status, then determines whether a cell should grow, shift toward repair, or initiate autophagy.

In other words, rapamycin was not merely an antifungal drug, nor merely an immunosuppressant.

It became a gateway into understanding how cells allocate and manage biological time itself.

In aging research, rapamycin created a major turning point. Animal studies suggested that lifespan could potentially be extended even when treatment began later in life, expanding the idea that biological aging may remain modifiable even after significant aging has already occurred.

However, this point must not be misunderstood.

Rapamycin is not a supplement.

Nor is it something to be casually used as an “anti-aging drug.”

It is a potent immunosuppressive medication with important risks, including infection, lipid metabolism abnormalities, oral ulcers, and delayed wound healing.

Nevertheless, the conceptual impact of this drug has been enormous.

Aging may not simply be the passive passage of time.

Rather, it is a biological process shaped by the complex interplay of nutrition, growth, repair, inflammation, immunity, and metabolism.

And perhaps some parts of that process can be decoded as signaling pathways.

When one reflects on it, there is something profoundly beautiful about the fact that beneath the silent gaze of the Moai statues, looking endlessly out toward the sea, there lay hidden a key to understanding the timing mechanisms of life itself.

Rapamycin was born from the soil of Rapa Nui, discovered as an antifungal agent, introduced clinically as an immunosuppressant, and eventually transformed aging research through the discovery of mTOR signaling.

It was truly a grand voyage in life science that began on a remote island.

Aging may not simply be fate.

It may be a signal.

And if it is a signal, then perhaps one day humanity will learn not only to read it, but also to tune it.


モアイチョコレート

今回のスタッフと患者さんへのお土産は、特注のモアイチョコレートにしました。

イースター島のあるレストランで、前の日にお願いすると、モアイの手作りチョコレートを作ってくれて、出発の空港まで持ってきてくれるのです。

僕が注文したのは茶色と白色のチョコレート。

帰国後、患者さんにお配りしていますが、好評でしたよ。

 


絶海の孤島に落ちる夕日

イースター島での神々しい日の入りです。

こうやって大自然の中で、ゆっくりと日が沈むのを見ていると、普段の生活と離れて、いろいろな考えが整理されてきます。

今年購入するレーザー機器を何にするか?

来年度のレーザー学会や皮膚科学会に向けた研究をどうするか?

などなど…。

南十字星

サザンクロス、南十字星ってあまり見ることができないですよね。

あまりよく写らなかったのですが、写真を撮りました。

この南半球でしか見れない、世界で一番小さな星座は、南半球の人たちにとって大切な意味を持っていました。

磁針を持たないポリネシアの人々は、小さなカヌーに乗って、

昼間は太陽の上がる場所を、

夜はこの星座を目安に航海を続け、

新たな島探しの旅に出たのです。

西暦500年にこの島に初めて滞在したポリネシア人も、この星座を頼りにやってきたのですね。


ポリネシアンショー

イースター島の二日目の夜に、ポリネシアンショーを見にゆきました。

このショーはチリで賞を取ったほど、クオリティーが高いものだと言われ、一時間ぐらいのショーを観たのですが、独特のリズムに乗ったポリネシアンダンスは面白かったですよ。
